Summary

Cytokeratin 5 is a basic protein that is closely related to cytokeratin 6. They share similar tissue distribution and are found in various proportions in many non-keratinizing stratified squamous epithelia eg. tongue mucosa, as well as in basal epithelia of trachea, basal cells of epidermis, hair follicles, sebaceous and sweat glands of skin, luminal cells of the mammary gland, basal cells of prostate, urothelium, vagina and endocervical mucosa. Cytokeratins 5 and 6 are also expressed in basal cell epitheliomas, squamous cell carcinomas of skin, tongue, epiglottis and of the rectal-anal region. Cytokeratin 5&6 is a useful aid for classification of epithelioid mesotheliomas.

Functions

  • Required for the formation of keratin intermediate filaments in the basal epidermis and maintenance of the skin barrier in response to mechanical stress (By similarity). Regulates the recruitment of Langerhans cells to the epidermis, potentially by modulation of the abundance of macrophage chemotactic cytokines, macrophage inflammatory cytokines and CTNND1 localization in keratinocytes (By similarity).

Known Applications & Suggested Dilutions

  • Immunohistochemistry (Formalin-fixed) (1-2ug/ml for 30 minutes at RT),(Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95&degC followed by cooling at RT for 20 minutes),Optimal dilution for a specific application should be determined.
